The Rune Knight belongs to the monster family Knights that can be found in Castle of Kings (combatArea).svg Castle of Kings.
Rune Gear can obtain by defeating the Rune Knight and also by Smithing.
He also drops the Amulet of Glory, which is currently the best Amulet for Combat when upgrated to the Elite Amulet of Glory.
For farming the Amulet you should concider Scattered Goo Monster instead, due to it's higher Drop Rate and lower Combat Level.
